Insulin pretreatment for 8, 16, 20, 24 hours resulted in reduction of bGH- induced phosphorylated Stat5B. Maximal inhibition (25-30% of control) was observed at all times between 12 and 24 hours pretreatment with insulin. Moreover Insulin pretreatment for 16 hours also significantly inhibited bGH induced Jak2 tyrosine phosphorylation. These results demonstrate that Growth hormone stimulated Jak2/Stat5B phosphorylation was inhibited by chronic insulin treatment and Jak2 mediates STA5B phosphorylation.
