Also U937 cells were treated with 1 nanomolar of TNFSF13B for various lengths of time to assay the kinetics of NFKB activation all of which indicated that TNFSF13B activates NFKB in a dose and time dependent manner. The gel shift band was specific as its formation could be eliminated with excess unlabeled oligonucleotide and was supershifted by anti-p50 antibody only. To further ascertain NFKB activation was due to proteinaceous nature of TNFSF13B, protein was either digested with trypsin or heat-denatured prior to treatment, both of which completely abolished NFKB activation confirming that TNFSF13B was responsible for this activation." ;
