This increase is attributable to the rise in cyp7a activity, which catalyzes the rate-limiting step of the classical pathway. Interrupting the return of bile acids to the liver, by diverting bile or by feeding a bile acid-binding resin, also stimulates the synthesis of bile acids. In contrast, reintroduction of bile acids into bile-diverted rats reverses the stimulatory effect, indicating that bile acid synthesis is subject to end-product inhibition. It was later discovered that cyp7a enzyme activity is closely correlated with cyp7a mRNA abundance, indicating that cyp7a gene transcription is the major determinant of cyp7a activity.
