Treatment of 50ng/ml NGF for 48 hours increased approximately four-fold APP promoter activity (this induction is mediated by DNA sequences located between the nucleotides -307 and -15). Incubation of PC-12 cells with NGF for 48 hours lead to an increase in the intracellular APP content. Treatment of PC-12 subclone M-M17-26 (which express the dominant inhibitory RAS mutant) with NGF was not able to stimulate the APP promoter activity, showing the requirement of the functional Ras for this response.
