PKD induces activation of MAP kinases, MEK4 and MEK3, 6 activities, which are upstream activators of JNK and p38, respectively. BMP-2 also induced the activation of these MEKs and this effect was blunted in AS-PKD (PKD1 antisense mRNA) compared with empty vector-transfected cells suggesting that PKD is probably influencing either the regulation or the activity of a MEK kinase. Thus, BMP2 activated p38 MAPk via PKD through MEK3/6.
